Comparison of Extracellular Vesicle Purification Methods using the ViewSizer 3000

Particle Characterization

Extracellular vesicles (EVs) are particles secreted by cells. It is an umbrella term to describe artificial cell-derived vesicles (ACDVs) or biogenesis-related vesicles such as exosomes, ectosomes, or microvesicles. EVs contain valuable information for both diagnostic and therapeutic purposes. However, the collection, storage, preparation, and separation of samples significantly influence EV yield both quantitatively and qualitatively.

To assess EV yield, particle size and particle number concentration stand as crucial parameters. While methods like transmission electron microscopy (TEM) and cryo-electron microscopy are employed for EV characterization, the sample preparation processes are complex and time consuming. Nanoparticle tracking analysis (NTA) offers an alternative, high-throughput technique for measuring the number concentration and particle size distribution. By leveraging simultaneous multi-laser NTA, we can effectively and efficiently evaluate various purification
